Introduction:

Eggshell membrane is composed of two individual membranes between egg albumin and eggshell. The membranes are primarily comprised of protein fibers. Eggshell membrane also has been shown to contain acid ~lycosaminoglycafls including dermatan sulfate and chondroitin-4-sulfate (*1). Picard, et.al., (*2) isolated and characterized sulfated glycoproteins from eggshell membrane. Glycoproteins included hexosamines, hexoses, and fucose. More recently, significant amounts of hyaluronic acid have been detected in eggshell membrane (Patent Pending, Membrell, LLC.).

Glucosamine and chondroitin sulfate have been purported to be effective in the treatment of osteoarthritis over the last 3 to 4 decades. (*3) Since eggshell membrane contains both glucosamine and chondroitin sulfate, it is suggested that this natural composition might be effective in relieving joint and soft tissue pain.

A single-site, open-label study was conducted to evaluate the effectiveness of Natural Eggshell Membrane (NEM) to relieve soft tissue and joint pain.

Study Parameters:

Eleven subjects with persistent pain were enrolled in an open-label evaluation of Natural Eggshell Membrane powder. Ten subjects, 5 male and 5 female, completed the 30-day follow-up. Subjects ingested one 500mg capsule of Natural Eggshell Membrane powder each day and rated their degree of pain using a 10-point analog pain assessment scale. Pain ratings were made before treatment, daily for 7-days, and at day 30. The clinician- investigator conducted range of motion measurements for both flexibility and extension before treatment, daily for 7-days, and at day-30. During range of motion measurements, subjects again rated the degree of pain.

Subject Population:

The population included subjects with lower back pain (4), knee (2), hip (1), shoulder (1), elbow (1) and cervical (1) pain. Pain etiology was predominately due to traumatic injury resulting from an accident or from surgery.

Study Results: 7-days

Pain assessment ratings, range of motion angles and pain associated with range of motion measurements were statistically evaluated using one-way analysis of variance and a means comparison analysis. Results showed no significant differences between the mean day-i pain ratings and the mean day-7 pain ratings. However, examination of individual pain ratings showed that 5 subjects had less pain at day-7, 3 subjects had more pain at day-7 and 2 subjects had the same pain at day-7.

Analysis of pain associated with range of motion tests was also conducted. The means comparison analysis showed that the average pain level associated with extension measurements was significantly less at day-7 compared to day-i. Pain from flexibility measurements was not significantly less at day-7 compared to day-i. However, the mean pain rating on day-7 was less than the mean pain rating on day-i. Overall examination of the pain ratings during range of motion measurements indicate that 4 subjects exhibited much less pain, 4 subjects exhibited slightly less pain and 2 subjects exhibited more pain.

Study Results: 30-days

A statistical comparison of day-30 mean pain ratings to day-I mean pain ratings showed that the day-30 pain ratings were significantly lower than the day-i pain ratings. NEM treatment resulted in a statistically significant reduction in pain at day-30 (mean = 1.1) compared to day-i (mean = 4.3).

A means comparison analysis showed that the average pain level associated with extension measurements was significantly less at day-30 compared to day-i. Pain from flex measurements was also significantly less at day-30 compared to day-i.

Conclusions:

Statistical analysis shows significant pain reduction (mean for all patients) after 30-days of treatment with Natural Eggshell Membrane powder. The results from this open-label study show that ingestion of one NEM capsule (500mg) each day for 30-days is effective in reducing pain resulting from a variety ofjoint and soft tissue insults.

In the second stage of the study all subjects received the same NEM composition. Mean pain scores continued to decrease in this stage. However, the differences between day-7 and day-30 pain assessment scores were not statistically significant, indicating that the primary reduction in pain occurred in the first 7-days.

Conclusions:

Statistical analysis shows significant pain reduction (mean for all patients) after 7-days of treatment with Natural Eggshell Membrane compositions. There were no significant differences between pain scores of subjects consuming the two different NEM compositions. Pain reduction continued during the second stage of the study, from day-8 through day-30. However, additional reductions in mean pain scores were not statistically significant. The results from this two-stage study show that ingestion of one NEM capsule (500mg) each day for a 7-day period is effective in reducing joint and soft tissue pain. Pain relief continues to improve with daily ingestion of NEM beyond 7-days.
